盆底器官脱垂患者主要支持韧带中雌激素受体GPR30表达的研究

Differential expression of estrogen receptor GPR30 in main supporting ligaments of patients with pelvic organ prolapsed#br#

摘要: 目的 · 研究盆底器官脱垂(POP)患者子宫圆韧带及主韧带中雌激素受体 GPR30 及支撑组织细胞外基质(ECM)主要成分 的表达情况。方法 · 选取 22 例 POP 患者(POP 组)和 10 例子宫颈上皮内瘤变Ⅲ级患者(对照组)作为研究对象。收集子宫切除标 本,采用 Western blotting 及免疫组织化学方法,对子宫圆韧带及主韧带组织中 GPR30 及弹性蛋白 (elastin)、Ⅰ型胶原蛋白(Col Ⅰ)、 Col Ⅲ的表达进行检测。结果 · Western blotting 检测及免疫组织化学评分结果均显示 POP 组患者子宫圆韧带及主韧带组织中 GPR30、 elastin、Col Ⅰ和 Col Ⅲ的表达显著低于对照组,差异均有统计学意义(P<0.05)。 结论 · POP 发生可能与相关组织中雌激素受体缺 乏、胶原蛋白的含量下降、elastin 减少有关。GPR30 是否参与盆底支撑组织 ECM 主要成分的合成、分泌及降解,有待进一步的深入 研究。

Abstract: Objective · To investigate the expression levels of estrogen receptor GPR30, elastin, collagen Ⅰ (Col Ⅰ), and Col Ⅲ in the uterus round ligament and the cardial ligament in patients with pelvic organ prolapse (POP) and main components in extracellular matrix (ECM).  Methods · 22 patients with POP (the POP group) and 10 patients with cervical intraepithelial neoplasia (CIN) Ⅲ (the control group) were enrolled. Hysterectomy specimens were collected and expressions of GPR30, elastin, Col Ⅰ and Col Ⅲ in the uterine round ligament and the cardial ligament tissues were detected with Western blotting and immunohistochemistry.  Results · Western blotting and immunohistochemistry results showed that the expressions of GPR30, elastin, Col Ⅰ and Col Ⅲ in the uterine round ligament and the cardial ligament tissues were significantly lower in the POP group than in the control group. The differences were statistically significant (P<0.05).  Conclusion · The occurrence of POP may be associated with estrogen receptor deficiency and decreased contents of collagen and elastin in relevant tissues. Whether GPR30 is involved in the synthesis, secretion and degradation of ECM major components in pelvic support tissues needs further investigation.
