下调 C2CD3通过抑制 Hedgehog通路调控上皮性卵巢癌 增殖、侵袭和迁移

Inhibition of C2CD3 regulates the proliferation, invasion and migration of epithelial ovarian cancer through inhibiting Hedgehog pathway

摘要: 目的 ·研究含 C2钙依赖性结构域蛋白 3(C2 calcium dependent domain containing protein 3,C2CD3)在卵巢癌组织中的表达与临床病理参数的关系,及其对卵巢癌 SKOV3细胞增殖、侵袭及迁移的影响和可能机制。方法 ·免疫组织化学方法检测卵巢癌组织及正常卵巢组织中 C2CD3的表达水平,分析 C2CD3表达水平与卵巢癌临床病理参数的相关性。下调 SKOV3细胞中 C2CD3基因, EdU检测细胞增殖能力, Transwell检测细胞侵袭能力,划痕实验检测细胞迁移能力, Western blotting检测 C2CD3、Shh、Ptch1、Smo及 Gli1蛋白水平。结果 · C2CD3表达于卵巢癌细胞质,在卵巢癌组织中 C2CD3呈强阳性表达,正常卵巢组织中呈弱阳性表达或不表达。与正常卵巢组织比较, C2CD3在卵巢癌中表达明显增高( P<0.05)。C2CD3表达与肿瘤分期及分级相关,分期越高,表达越强(P<0.05);分级越高,表达越强( P<0.05)。在不同年龄及组织类型中表达差异无统计学意义。下调卵巢癌 SKOV3细胞 C2CD3基因后,卵巢癌细胞增殖、侵袭和迁移能力均降低(均 P<0.05);C2CD3、Shh、Ptch1、Smo及 Gli1蛋白水平均下降。结论 · C2CD3在卵巢癌组织中表达上升。下调 C2CD3基因后,卵巢癌 SKOV3细胞增殖、侵袭和迁移能力均下降,可能与抑制 Hedgehog通路中 Shh、 Ptch1、Smo及 Gli1蛋白相关。

Abstract:

Objective &middot; To explore the relationship between the of C2 calcium dependent domain containing protein 3 (C2CD3) in ovarian cancer and clinicopathological parameters, and its effect on the proliferation and invasion of ovarian cancer SKOV3 cells and possible mechanisms. Methods &middot; The of C2CD3 protein in ovarian tissue was detectedimmunohistochemistry. The proliferation, migration and invasion abilities of SKOV3 cells were detectedEdU assay, wound healing assay and Transwell assay respectively. Western blotting was performed to investigate the of C2CD3, Shh, Ptch1, Smo and Gli1 proteins. Results &middot; C2CD3 protein was located in cytoplasm in ovarian cancer. C2CD3 was highly expressed in ovarian cancer compared to normal ovarian tissue. C2CD3 immunostaining was significantly higher in tumor samples in advanced stages (stage Ⅲ / Ⅳ ) than in early stages (stage Ⅰ/ Ⅱ ). The staining intensity was significantly correlated with the grade (grade3 vs. grade1+2). The association between C2CD3 and age (or tumor type) was not significant. Inhibition of C2CD3 gene significantly weakened the proliferation, invasion and migration abilities of SKOV3 cells, and the of Shh, Ptch1, Smo and Gli1 proteins was significantly decreased. Conclusion &middot; The of C2CD3 protein is increased in ovarian cancer tissues. Inhibition of C2CD3 gene weakens the proliferation, invasion and migration capacities of ovarian cancer SKOV3 cells, which may be related to the inhibition of Shh, Ptch1, Smo and Gli1 proteins in Hedgehog pathway.
