预防性给予姜黄素对大鼠溃疡性结肠炎病变及色氨酸羟化酶表达的影响

摘要:

目的 ·研究姜黄素对溃疡性结肠炎(UC)的预防作用及对色氨酸羟化酶(TPH)表达的影响。方法 ·使用5%多聚葡酸钠(DSS)诱导SD大鼠UC模型(连续饮用7 d),自造模第1日开始预防性给予姜黄素灌胃(60 mg/kg,连续10 d),记录大鼠疾病活动指数(DAI)评分。应用苏木精-伊红(H-E)染色方法对结肠进行组织学评分,评价姜黄素对炎症的影响;应用免疫印迹(Western blotting)方法检测姜黄素对炎症结肠组织5-羟色胺(5-HT)合成限速酶TPH表达的影响。结果 · DSS引起大鼠结肠组织出现UC组织病理学改变,DAI评分上升,TPH蛋白水平明显升高(P=0.000)。与对照组比较,预防性给予姜黄素能减轻结肠炎症组织学评分
(P=0.034),可降低DAI评分(P=0.045);但预防性给予姜黄素对炎症结肠组织中TPH的表达无影响。结论 ·预防性给予姜黄素可以抑制炎症反应,但不能抑制TPH表达上调,提示姜黄素的抗炎效应并不是通过抑制5-HT合成实现的。

Abstract:

Objective · To investigate the preventive effects of curcumin on inflammation and expression of tryptophan hydroxylase (TPH) in a rat model of ulcerative colitis(UC). Methods · UC was induced by 5% dextran sulfate sodium (DSS) in drinking water provided to SD rats for continuous 7 d .The oral administrations of curcumin (60 mg/kg, once a day for 10 d) were started at Day 1 since the establishment of UC model as preventive treatment. The anti-inflammatory effects of curcumin were assessed by disease activities index (DAI) score and colonic histological score(H-E staining). The effect of curcumin on expression of TPH was examined by Western blotting. Results · DSS-induced histological change in colon defined as UC increased in DAI score and increased in protein level of TPH in colon (P=0.000). Compared with the control group, preventive treatment with curcumin significantly inhibited histological score (P=0.034)and DAI score (P=0.045), while curcumin did not alter the DSS-induced upregulation of TPH. Conclusion · Preventive treatment with curcumin suppresses the inflammatory responses, while curcumin does not alter the DSS-induced upregulation of TPH. These findings indicate that anti-inflammatory effect of curcumin is independent of inhibition of 5-HT synthesis.
