HPV16 E2对宫颈癌SiHa细胞上皮间质转化的影响

摘要:

目的 探讨HPV16 E2对宫颈癌SiHa细胞上皮间充质细胞转化的影响。方法 采用脂质体转染p-CAG-MYC-HPV16 E2至宫颈癌SiHa细胞中,倒置相差显微镜下观察转染前后细胞形态的改变;RT-PCR及Western blotting检测转染前后Twist2、上皮细胞标志物E-cadherin及间质细胞标志物Vimentin表达的变化。结果 成功转染HPV16 E2至宫颈癌SiHa细胞中。转染后,上皮细胞由椭圆形变成梭形,细胞间隙增大;Twist2和Vimentin表达上调,E-cadherin表达下调。结论 HPV16 E2能在体外诱导宫颈癌细胞发生上皮间充质细胞转化,促进早期癌变。

Abstract:

Objective To investigate the influence of HPV16 E2 on epithelial-mesenchymal transition of cervical cancer SiHa cells. Methods Lipofectamine was used to transfect p-CAG-MYC-HPV16 E2 gene into cervical cancer SiHa cells. The morphology of cells was observed before and after transfection with phase-contrast microscope, and the expression of Twist2, E-cadherin and Vimentin before and after transfection was determined by RT-PCR and Western blotting. Results HPV16 E2 was successfully transfected into SiHa cells. After transfection, the epithelial cells became spindle-shaped, with enlarged intercellular space, and the expression of Twist2 and Vimentin was up-regulated, while the expression of E-cadherin was down-regulated. Conclusion HPV16 E2 can induce epithelialmesenchymal transition of cervical cancer cells, and promote cervical carcinogenesis.
