Forkhead transcription factor Foxa2 is an important transcription factor regulating pancreatic development and glucose and lipid metabolism. As a downstream target of insulin signaling, constitutive activation of Foxa2 in the brain results in increased food consumption, metabolism and insulin sensitivity. Foxa2 regulates gene expression involved in glucose and lipid metabolism pathway, improves insulin resistance in peripheral tissues, and has been recognized as a novel transcription factor of insulin sensitivity. Phosphorylation of the protein results in Foxa2's inactivation by its localization changing from nuclei to cytosol. The research progress of relationship between Foxa2 and glucose and lipid metabolism and diabetes mellitus is reviewed in this paper.
