Preliminary study of flavonoids affecting cAMP level in cannabinoid receptor overexpressed cells#br#

Abstract: Objective · To evaluate the effects of flavonoids on cyclic adenosine monophosphate (cAMP) level in cannabinoid receptor (CBR) overexpressed cells.  Methods · Eukaryotic expression vectors GV144-CB1R and GV144-CB2R were constructed, and transfected into HEK-293 cells to establish CBR overexpressed cell lines. Several flavonoid compounds were studied by measuring the change of cAMP level in CBR-overpressed HEK293 cell lines.  Results · The CBR overexpressed cell lines were constructed successfully. cAMP level in CBR-overexpressed HEK-293 cells decreased after incubation with flavonoid compounds. And these effects were selective for CB2R overexpressed cells.  Conclusion · All flavonoids studied exhibited decreasing effect on cAMP level in CBR-overexpressed cells at μmol/L level.
