Influence of HPV16 E2 on epithelial-mesenchymal transition of cervical cancer SiHa cells

Abstract:

Objective To investigate the influence of HPV16 E2 on epithelial-mesenchymal transition of cervical cancer SiHa cells. Methods Lipofectamine was used to transfect p-CAG-MYC-HPV16 E2 gene into cervical cancer SiHa cells. The morphology of cells was observed before and after transfection with phase-contrast microscope, and the expression of Twist2, E-cadherin and Vimentin before and after transfection was determined by RT-PCR and Western blotting. Results HPV16 E2 was successfully transfected into SiHa cells. After transfection, the epithelial cells became spindle-shaped, with enlarged intercellular space, and the expression of Twist2 and Vimentin was up-regulated, while the expression of E-cadherin was down-regulated. Conclusion HPV16 E2 can induce epithelialmesenchymal transition of cervical cancer cells, and promote cervical carcinogenesis.
