ID1蛋白在胃癌组织中的表达及临床意义

Expression and clinical significance of ID1 in gastric cancer

摘要:

目的 分析人胃癌组织中分化抑制因子1 (ID1)的表达与临床病理参数之间的关系,初步探讨ID1在胃癌发生、发展及患者预后评估中的作用。方法 免疫组织化学EliVision法检测91例胃癌组织、30例正常胃黏膜和35例不典型增生胃黏膜组织中ID1蛋白的表达,分析ID1蛋白表达与胃癌的分化程度、浸润深度、临床TNM分期和淋巴结转移的关系,绘制生存曲线,Log-rank法单因素分析肿瘤临床病理学特征和ID1蛋白的表达与患者预后的关系。结果 免疫组织化学染色结果显示:胃癌组织中ID1蛋白的表达阳性率为63.7% (58/91),显著高于正常胃黏膜组织(10.0%)和不典型增生胃黏膜组织(42.9%),差异均有统计学意义(P<0.05)。统计学分析结果显示,胃癌组织中ID1蛋白的表达与肿瘤的分化程度、浸润深度、临床TNM分期和淋巴结转移有关(P<0.05)。Log-rank法单因素分析结果显示,肿瘤浸润深度与淋巴结转移与胃癌患者的预后有关(P<0.05),而与患者年龄、性别及肿瘤直径、大体类型、分化程度、Lauren分型和ID1蛋白的表达等无关(P>0.05)。结论 ID1在胃癌组织中表达异常,可能与胃癌的发生和发展有关,有望为胃癌治疗提供新的治疗靶点,但并非是胃癌患者预后的评估指标。

Abstract:

Objective To investigate the relationship between expression of inhibitor of DNA binding/differentiation 1 (ID1) and clinicopathological parameters, and explore the role of ID1 in the development and prognosis prediction of gastric cancer. Methods The expression of ID1 protein in 91 samples of gastric cancer tissues, 30 samples of normal gastric tissues and 35 samples of atypical hyperplasia gastric tissues was examined by immunohistochemical EliVision method. The relationship between expression of ID1 protein and differentiation, invasion, TNM stage and lymph node metastasis of gastric cancer was analysed, the survival curves were drawn, and the correlation of clinicopathological parameters and expression of ID1 protein with prognosis of patients with gastric cancer was explored with Log-rank univariate analysis. Results Immunohistochemical staining revealed that the positive rate of expression of ID1 protein in gastric cancer tissues was 63.7% (58/91), which was significantly higher than those in normal gastric tissues (10.0%) and atypical hyperplasia gastric tissues (42.9%) (P<0.05). Statistical analysis indicated that the expression of ID1 protein was related to the differentiation, invasion, TNM stage and lymph node metastasis of gastric cancer (P<0.05). Log-rank univariate analysis demonstrated that the invasion and lymph node metastasis of gastric cancer were related to the prognosis of patients (P<0.05), while the prognosis of patients was not related to the age, sex, diameter of tumor, gross type, differentiation, Lauren classification and expression of ID1 protein (P>0.05). Conclusion The expression of ID1 in gastric cancer is abnormal, which may be associated with  the occurrence and development of gastric cancer. ID1 may be the new therapeutic targets for the treatment of gastric cancer, while it may not be the prognostic factors of patients with gastric cancer.
