Effects of hypoxia culture on damage and apoptosis of RL95-2 cell line

Abstract:

Objective To investigate effects of hypoxia culture on mitochondria damage and apoptosis of human endometrial epithelial carcinoma cell line (RL95-2). Methods RL95-2 cells were cultured with medium containing different concentrations of CoCl2 (0, 10, 30, 62.5, and 125 μmol/L). Cell proliferation activity, mitochondria membrane potential, and apoptosis rate were determined via CCK-8 kit, JC-1 fluorescence staining with flow cytometry, and Annex V/PI. Results With CoCl2 concentration rising or extension of action time (62.5 μmol/L), the proliferation activity of RL95-2 cells was weak, while the depolarization of mitochondria membrane and the apoptosis rate were increased. Conclusion The condition of hypoxia induces decrease of proliferation activity, mitochondria damage, increase of apoptosis of endometrial epithelial cells.
