滤泡性辅助 T 细胞亚型与原因不明复发性流产的关系研究

Relationship between subtypes of T follicular helper cells and unexplained recurrent spontaneous abortion

摘要: 目的 · 探讨原因不明复发性流产(URSA)的发生与滤泡性辅助T 细胞(Tfh)亚型的关系。方法 · 以 28 例正常人工流产 患者为对照组,28 例染色体正常的URSA 患者为流产组。分离2 组患者外周血及子宫蜕膜组织中单个核细胞,用流式细胞术检测 CXCR3+CCR6-、CXCR3-CCR6-及 CXCR3-CCR6+ 3 种 Tfh 细胞亚型水平及B 细胞水平。结果 · 流产组蜕膜组织CXCR3-CCR6- Tfh 细胞水平显著高于对照组(P=0.015),且与 B 细胞水平有相关性(R2=0.779, P=0.025)。 2 组蜕膜组织中另外 2 种 Tfh 细胞亚型水 平以及外周血中 3 种 Tfh 细胞亚型水平差异均无统计学意义(均 P>0.05)。 结论 · 早孕妇女蜕膜组织局部 CXCR3-CCR6- Tfh 细胞亚 型数量增多可能与 URSA 发生有关。

Abstract:

 Objective · To discuss the relationship between unexplained recurrent spontaneous abortion (URSA) and T follicular helper (Tfh) cell subtypes.  Methods · Twenty-eight normal early pregnancy women, who had undergone induced abortion, were taken as control, and 28 patients with URSA were enrolled in the abortion group. The mononuclear cells in peripheral blood and decidual tissues were separated in the two groups, and CXCR3+CCR6- Tfh cells, CXCR3-CCR6- Tfh cells, CXCR3-CCR6+ Tfh cells and B cells were tested by flow cytometry.  Results · The decidual CXCR3CCR6- Tfh cells significantly increased in the abortion group compared with control group (P=0.015). And there was a strong association between the decidual CXCR3-CCR6- Tfh cells and B cells in URSA patients (R2=0.779, P=0.025).  Conclusion · The up-regulation of decidual CXCR3-CCR6- Tfh cells in early pregnancy women may be related with the occurrence of URSA.
