老年心肌梗死患者外周血全基因组DNA甲基化和羟甲基化水平与冠状动脉粥样硬化程度的关系

Relationship between global DNA methylation and hydroxymethylation levels in peripheral blood of elderly patients with myocardial infarction and the degree of coronary atherosclerosis

摘要: 目的·探讨老年心肌梗死(myocardial infarction,MI)患者外周血中全基因组DNA甲基化和羟甲基化的水平与冠状动脉粥样硬化程度的关系。方法·收集43例确诊为MI的老年患者为研究组,同期53例冠状动脉造影检查阴性的老年患者为对照组。采集患者外周血,酶联免疫法测定外周血全基因组DNA甲基化和羟甲基化水平以及血浆中甲基双加氧酶(ten-eleven translocation-2,TET2)浓度,实时荧光定量PCR测定TET2 mRNA表达水平。结果·与对照组相比,MI患者外周血全基因组DNA甲基化和羟甲基化水平显著增加(P0.014,P0.000),血浆TET2浓度及其mRNA表达水平显著增加(P0.004,P0.028);DNA甲基化和羟甲基化水平与冠状动脉粥样硬化程度显著相关(r0.262,P0.017;r0.360,P0.001)。结论·老年MI患者外周血中全基因组DNA甲基化和羟甲基化的水平增加,其与冠状动脉粥样硬化程度显著相关。

Abstract:

Objective · To investigate the relationship between the levels of global DNA methylation and hydroxymethylation in peripheral blood of elderly patients with myocardial infarction (MI) and the degree of coronary atherosclerosis. Methods · Forty-three elderly patients with MI were enrolled as the study group and 53 elderly patients with negative findings in coronary angiograms during the same period were collected as the control group. Peripheral blood samples were collected. The levels of global DNA methylation and hydroxymethylation in peripheral blood and the concentration of plasma TET2 were detectedELISA. TET2 mRNA was determinedreal-time qPCR. Results · Compared with the control group, the levels of global DNA methylation and hydroxymethylation in peripheral blood of elderly patients with MI were significantly increased (P0.014, P0.000). The concentration of plasma TET2 and its mRNA in patients with MI were significantly higher than those in the control group (P0.004, P0.028).The levels of global DNA methylation and hydroxymethylation in MI group were significantly correlated with the degree of coronary atherosclerosis (r0.262, P0.017; r0.360, P0.001). Conclusion · The levels of global DNA methylation and hydroxymethylation in peripheral blood of elderly patients with MI are significantly increased, which are significantly correlated with the degree of coronary atherosclerosis.
